Little details on larger diatoms (more 365nm UV images)

I imaged a few larger and/or thicker diatoms with 365nm light to test the limits of my current UV setup. I used an Oly UPlanSApo 100/1.4 on an Oly BX61 for all images. Overall results came out a little better than I expected, which was nice.

1. Trigonium articum var pentagonalis. A large diatom that almost filled the FoV but isn't very thick (~25µm). The detail isn't that small on this one either, but it rendered well.

2.Brightwellia coronata. A smaller form with easy to resolve details but thicker than T. articum (35µm+). It rendered nicely too

3. Aulacodiscus cf angulatus. A thick specimen (~45µm) with lots of fine details, some in separate but closely-spaced layers in the stack. Clarity drops off in deeper parts nearer the edge of the form, but that's expected. Overall it's still better resolution than white or even blue light would produce under the same conditions.

4. Aulacodiscus sollittianus. The three long "horns" (ocelli) touching the underside of the coverslip pushed the rest of the form very deep into the mount (~55-60µm). Even so, some pretty fine detail away from the edges was successfully resolved. Happy with that.

Note: All images were downsampled to 2k square. The insets were clipped from the original, full-sized images and then pasted into the downsamples. They show a 100% pixel-for-pixel view of captured detail (when the main 2k image is viewed full size on screen).

Thanks for looking

The best light microscopy images of diatoms I have seen so far. Congratulations on these images.

Dear Steve,
You've still got a surprise up your sleeve. Amazing photos!!!

You're the master of image resolution.
