Wet mounts - how to?

I would ask if any of you have any experience making wet mounts and would share it with me.

I have got several very interesting plankton samples from around the world with a lot of very weird creatures in and many too big, to make ordinary slides of. Recently I swapped some very interesting samples of zooplankton from the north Atlantic Sea near the Faroe Islands. It was a university department that was not able to find small glass pearls and I swapped some of my pearls for three big professional samples.

    

I have – for several years - tried to make wet mounts using ordinary all. rings and special rings with creatures preserved in alcohol and formalin but have not been able to seal them tight.

I have used the method described in: Grey: The Microtomist formulary and used goldsize, diff. Two components et cet, but not been able to get them tight.



For some weeks ago someone wrote of a source of asphalt varnish, which I have tried to get for along time. I have just got it this week, but not tried it as yet. I am planning to try this next. Else I will try to move the specimens from alcohol/formaldehyde to glycerine or glyceringelatine, as it should not vaporize as easily as alchol/formaldehyde.

So if anyone have a foolproof method – I would very much like hear about it – preferably just in German.

Hallo Klaus,
In england gibt es eine Firma die Aquamount liefert.
Wasserhaltige Objekten können soft mit diesem Mittel eingedeckt werden.
Mann braucht das Umranden auch nicht bei dieses Material.
Die Firma ist NBS.
Hoffentlich habe ich Dich etwas geholfen.

Thanks for the tip. I have read about it but not tried it. My main reason wanting to make wet mounts is that the bigger critters is about 2-3 mm thick and that is a problem with ordinary mountants even with glass pearls. But if you have experience with Aquamount and have slides with such thick layers, I will certainly try it out. If I remember right it is rather expensive. I have just tried Google it, but didnt find any suppliers. I know NBS and have all their booklets, but I thought It was closed several years ago when Marson died?

Hallo Klaus,

ich mache solche Präparate wie folgt.

Ich benutze Fiber - Ringe aus dem Sanitärfachhandel mit einem Innendurchmesser mit 15 mm und einem Außendurchmesser mit 19 mm.

Diese Ringe klebe ich mit Sekundenkleber auf einen Objektträger. Die entstehende Höhlung streiche ich vollständig dünn mit einem Einschlussmittel aus ( zur Abdichtung) und lasse trocknen.  Ich füge dann meine einzuschließende Flüssigkeit ein, dabei halte ich zum oberen Rand etwa einen Abstand von einem halben bis einem mm ein. Ich lege dann auf diese Flüssigkeit im inneren des Ringes ein rundes Deckglas mit einem Durchmesser von 14 mm auf, so daß es schwimmt/ sich am Rand verkeilt. Dabei darf keine Flüssigkeit auf die Oberseite des Deckglases gelangen. Zwischen Deckglas und Flüssigkeit sollen auch sich keine Luftblasen befinden was bei sehr sauberen Deckgläsern garnicht so schwer ist. Auf dieses Deckglas gebe ich 2- 3 Tropfen eines zähen Einschlussmittels, mit schnellen Trockungzeiten z.B. Entellan. Durch die Zähigkeit läuft es- wenn alles gut geht -  nicht in den Flüssigkeitsbereich hinein und härtet sehr schnell aus. Manchmal kippt aber auch das Deckglas oder die Verkeilung war nicht fest genug, dann ist das Präparat verdorben.  Falls es klappt folgt ein weiterer Tropfen, der dann den Hohlraum bis zum Rand ausfüllt und mit dem Abdeckdeckglas von 18 mm Durchmesser sauber abschließt.

That sounds as a new and very interesting method, but aso one requiring a very stable hand? I would be very glad if you had some pictures of your finished slides and if possible of the intermediate steps.

To what purpose do You use those slides? Also plankton?

Schau mal hier oben, dort gibt es links auch NBS.
Ich weiss nicht mehr ob es teuer war ich habe es schon lange und benutze es für das einschliessen von Radiolarien und Foraminiferen.
Ich benutze ringen von 2-3 mm hoog, durchmesser 18mm, damit ich deckgläser von 18 mm gebrauchen kann zum abdecken.
die Foraminiferen werden auf eine Unterlage geklebt mit Tragacanth und dann wird der Kammer aufgefühlt mit Aquamount, Deckglas darürber und fertig ist dein Präparat.

Thanks for the comments. Is it Brunnel You meant and their Aqueous mountant? Tragacanth I have. So my idea with glyceringelatine should work also.
