Was benötigt man, um ein Zeiss Standard auf DIC/DIK umzurüsten?

Hi there,

I think René is right on this one. The objective will not change from NA 1.4 to 0.99; is stays at 1.4.

When using an immersion condenser NA=1.4 without immersion, the aperture will be somewhere between 0.8 and 0.9 (depends on the model; hard to predict because the condenser will usually not be corrected for both modes, but not 0.99).

When using an objective NA=1.4 with a condenser NA=1.4, the maximum resolution that can be achived is 240 nm (for green light). When the same objective is used with the dry condenser NA=0.9, the resolution will be 270 nm; at NA=0.8, the resolution is 277 nm.

The calculations are from this publication:
Baker (1952): Remarks on the Effect of the Aperture of the Condenser on Resolution by the Microscope. Quarterly Journal of Microscopical Science 93: 375-377.

Kind regards,

Jon

Hello René!

As an aside, in DIC mode, the effect is less because of the full aperture lighting that is generally employed: the condenser iris is not closed to near the phase ring.

Well, that's good to hear. However, even if phase rings will not lead to any distortions, I guess that it would be still necessary to use Plan-objectives in DIC mode, isn't it?

kind regards,
Michael

Hello Michael,

Well, the old Zeiss DIC was calculated to be used with planachro's, but there are no principal reasons for that.

Good luck hunting, René

All,
